Fluorescence Spectrophotometer - Agilent Technologies Cary Eclipse

The Cary Eclipse Spectrophotometer uses a Xenon flash lamp to measure the emission of light from samples in four modes. With superior sensitivity, high signal-to-noise ratio and fast kinetics, it captures a data point every 12.5 ms and scans at 24,000 nm/min without peak shifts.

  • Fluorescence, phosphorescence, chemiluminescence and bioluminescence modes
  • Light source: Xenon flash lamp
  • Lamp pulse width at half peak height ~2 µs, peak power equivalent to 75 kW
  • Limit of detection:
    • 1.0 picomolar fluorescein (< 500 µL sample volume) using a standard 10 mm cuvette
    • 10 picomolar fluorescein (< 40 µL sample volume) using a 40 µL micro-cuvette
  • Separate monochromators for excitation and emission
  • Wavelength range: 200 – 900 nm for both excitation and emission monochromators
  • Wavelength accuracy: ±0.5 nm at 541.2 nm and ±1.0 nm from 200 – 900 nm
  • Wavelength reproducibility: ±0.2 nm
  • Spectral band pass - Excitation: 1.5, 2.5, 5, 10, 20 nm; Emission: 1.5, 2.5, 5, 10, 20 nm
  • High sensitivity: red-sensitive photomultiplier tube detectors extend to 900 nm
  • Fast kinetics: scan speed of 24,000 nm/min without peak shifts and up to 80 data points per second in steady-state fluorescence mode
  • Data point capture capability:
    • One phosphorescence data point every 1 µs up to 10 s
    • One fluorescence data point every 12.5 ms up to 999 s
    • One bio/chemiluminescence data point every 40 µs up to 10 s
  • Build-in excitation and emission filters to exclude secondary light
  • Room light immunity for most fluorescent samples
  • Cary Eclipse Control Software
